Immunohistochemical Analysis of Glutathione Transferase Isoenzymes in the Rat Muscle Tissue: a specific marker for type II muscle fiber


Abstract
Glutathione transferases (GST) are the enzymes mainly responsible for the biochemical detoxification by conjugating glutathione to a set of ligands. The isoenzyme pattern of the enzymes had been elucidated. But in case of muscle tissues, the
studies in
relation with GST have not been done thoroughly. In this experiment, we carried out the immunohistochemical analysis on the distributin of GST isoenzymes using anti GST-Pi and GST-L antibodies on the cardiac, smooth and skeletal muscle tissues.
The
results showed that the intestinal smooth muscle and cardiac muscle were very weakly or negatively responsive to both of anti GST-L and Pi antibodies. However, the skeletal muscle did no react with anti GST-Pi antibody, but the major bundles of
skeletal
muscle were positively responsive to GST-L antibody. Therefore, we compared the expression of GST-L in the muscle tissues of soleus and plantaris, which composed predominantly of type I fiber and type II fiber, respectively. The data indicated
that
only, some of type II fiver in the plantaris muscle tissue were positive to GST-L antibody, which was confirmed by specific ATP ase staining. Therefore, it can be suggested that the expression of GST isoenzyme can be used as a type-specific
marker
for
type II fiber of skeletal muscles.
